Britanski znanstvenici razvili su novi alat kojim iskorištavaju stanice koje uništavaju tumore za brzu i učinkovitu borbu protiv raka.
Nova studija pokazala je dramatično smanjenje tumora kod miševa u usporedbi s tradicionalnim metodama stanične terapije koje poboljšavaju sposobnost imunosnog sustava u borbi protiv raka.
S novim uređajem koji bi se mogao stvoriti uz pomoć 3D printera, znanstvenici sa Sveučilišta Northwestern umnožili su, sortirali i sakupili stotine milijuna stanica, dobijajući 400 posto više stanica koje uništavaju tumore od dosadašnjih pristupa liječenju.
Većina metoda liječenja raka uključuje otrovne kemikalije i strane tvari, koje uzrokuju štetne nuspojave i slabe imuni odgovor tijela.
Korištenje vlastitog tkiva i stanica može eliminirati nuspojave i rizik od odbacivanja.
Shana Kelley, koja je sudjelovala u istraživanju, rekla je: "Ljudi su izliječeni od uznapredovalog melanoma kroz liječenje vlastitim imunosnim stanicama koje su sakupljene iz tumorskog tkiva".
"Problem je u tome što zbog načina na koji se stanice prikupljaju, to djeluje samo kod vrlo malog broja pacijenata."
Te stanice, nazvane limfociti koji infiltriraju tumor (TIL), prirodne su imunosne stanice koje napadaju tumorsko tkivo angažirajući stanice da se 'bore prsa u prsa', kažu znanstvenici.
Prema znanstvenicima stanične terapije koje se danas koriste u klinikama koriste mješavinu "istrošenih" i "nezrelih" stanica za liječenje tumora.
Nakon što se izvade iz tkiva, stanice se uzgajaju u laboratorijima daleko od pacijenata od kojih su uzete.
Do trenutka kada su se umnožile i spremne su za povratak u tijelo, mnoge stanice su iscrpljene i nesposobne se boriti, budući da su predugo u tumoru.
No, koristeći novu tehnologiju zvanu mikrofluidno ciljanje afiniteta infiltrirajućih stanica (MATIC), znanstvenici mogu točno odrediti koje su najaktivnije stanice.
U radu su znanstvenici koristili MATIC kako bi pronašli ciljanu populaciju stanica za miševe koje su proučavali.
Otkrili su da su se tumori kod životinja dramatično smanjili - a kod nekih miševa potpuno su nestali, što je dovelo do velikog poboljšanja stope preživljavanja u usporedbi s tradicionalnijim metodama.
Dr Kelley je rekla: “Umjesto da miševima damo mješavinu stanica s različitim fenotipovima, mi im dajemo fenotip jedne stanice koji im zapravo može pomoći".

Britanski znanstvenici razvili su novi alat kojim iskorištavaju stanice koje uništavaju tumore za brzu i učinkovitu borbu protiv raka.
Nova studija pokazala je dramatično smanjenje tumora kod miševa u usporedbi s tradicionalnim metodama stanične terapije koje poboljšavaju sposobnost imunosnog sustava u borbi protiv raka.
S novim uređajem koji bi se mogao stvoriti uz pomoć 3D printera, znanstvenici sa Sveučilišta Northwestern umnožili su, sortirali i sakupili stotine milijuna stanica, dobijajući 400 posto više stanica koje uništavaju tumore od dosadašnjih pristupa liječenju.
Većina metoda liječenja raka uključuje otrovne kemikalije i strane tvari, koje uzrokuju štetne nuspojave i slabe imuni odgovor tijela.
Korištenje vlastitog tkiva i stanica može eliminirati nuspojave i rizik od odbacivanja.
Shana Kelley, koja je sudjelovala u istraživanju, rekla je: "Ljudi su izliječeni od uznapredovalog melanoma kroz liječenje vlastitim imunosnim stanicama koje su sakupljene iz tumorskog tkiva".
"Problem je u tome što zbog načina na koji se stanice prikupljaju, to djeluje samo kod vrlo malog broja pacijenata."
Te stanice, nazvane limfociti koji infiltriraju tumor (TIL), prirodne su imunosne stanice koje napadaju tumorsko tkivo angažirajući stanice da se 'bore prsa u prsa', kažu znanstvenici.
Prema znanstvenicima stanične terapije koje se danas koriste u klinikama koriste mješavinu "istrošenih" i "nezrelih" stanica za liječenje tumora.
Nakon što se izvade iz tkiva, stanice se uzgajaju u laboratorijima daleko od pacijenata od kojih su uzete.
Do trenutka kada su se umnožile i spremne su za povratak u tijelo, mnoge stanice su iscrpljene i nesposobne se boriti, budući da su predugo u tumoru.
No, koristeći novu tehnologiju zvanu mikrofluidno ciljanje afiniteta infiltrirajućih stanica (MATIC), znanstvenici mogu točno odrediti koje su najaktivnije stanice.
U radu su znanstvenici koristili MATIC kako bi pronašli ciljanu populaciju stanica za miševe koje su proučavali.
Otkrili su da su se tumori kod životinja dramatično smanjili - a kod nekih miševa potpuno su nestali, što je dovelo do velikog poboljšanja stope preživljavanja u usporedbi s tradicionalnijim metodama.
Dr Kelley je rekla: “Umjesto da miševima damo mješavinu stanica s različitim fenotipovima, mi im dajemo fenotip jedne stanice koji im zapravo može pomoći".
"Uočite mnogo snažniju i mnogo veću stopu odgovora kada stvarno izlučite ciljanu reaktivnost T stanica."
Rezultati istraživanja objavljeni su u časopisu Nature Biomedical Engineering.
